Glutathione
Glutathione (GSH, Glu-Cys-Gly, CAS 70-18-8) is the dominant intracellular low-molecular-weight thiol, synthesized de novo in virtually all mammalian cell types via the gamma-glutamylcysteine synthetase pathway. Three enzyme-class interactions define its research relevance: it is the obligate cofactor for glutathione peroxidase-mediated peroxide reduction, the electrophile-conjugating substrate for glutathione S-transferases in phase II xenobiotic metabolism, and the reductant in the glutathione-ascorbate cycle that regenerates oxidized ascorbate. In research applications it is used to impose defined thiol/disulfide conditions in cell-free redox assays and to investigate thiol-exchange chemistry in protein biochemistry.
